PHARMACOKINETICS OF NIFEDIPINE AND PREDICTION OF PLASMA CONCENTRATION ON ESSENTIAL HYPERTENSIVE PATIENTS

The pharmacokinetics after oral administration of nifedipine have been studied in ten healthy male volunteers. The plasma concentration-time course pattern of nifedipine was quite liable to variation within individual subjects and a great variation of absorption rate constant was observed from the results of pharmacokinetic analysis. A program which could predict the blood level curve after repetitive dosing of various dose sizes at non-uniform dosing intervals was developed, and the prediction of plasma levels after multiple dosing of nifedipine was attempted on three essential hypertensive patients by using this program. The predicted plasma levels were in good agreement with observed plasma levels; therefore this program is suited to designing optimum dosing schedules for individual patients. A significant correlation between the plasma concentration of nifedipine and the change in systolic and diastolic blood pressure reduction was observed.
