I investigated the effects of cytosine arabinoside (araC) on single-strand break (SSB) repair, repair replication and recovery of sublethal damage (SLD) in X-irradiated HeLa S3 cells. The most effective araC treatment was the protocol involving addition and presence of araC (10^-5 M) before (30 minutes) and for various lengths of time after X-irradiation of the cells. AraC partially inhibited SSB repair and repair replication after 20 krad, and almost completely SLD recovery after relatively low X-ray doses, strongly suggesting a new finding that SSB repair requiring repair replication at the molecular level may be at least in part responsible for SLD recovery at the cellular level. It seems that this result will shed further light on the combined chemoradiotherapy of cancers, especially using low LET radiation.
