抗うつ剤の臨床的効果は主としてモノアミン取り込み抑制効果によると考えられてきたが,躁鬱病においてGABA神経伝達の関与する可能性が幾つかの実験によって示唆されてきた。11種類の三環系,四環系,四環系抗うつ薬がCOS-7細胞に発現させたGABAトランスポーターのサブタイプ(GAT1,GAT2,GAT3) ,セロトニントランスポーター(SET),グルタミン酸トランスポーター(GLT-1)に与える影響について検討した。実験に用いた抗うつ剤のすべてがセロトニントランスポーターの活性を濃度依存性に抑制した。desipramine,nortriptyline, amitriptyline, maprotilineはGABAトランスポーターの幾つかのサプタイプを抑制した。amitriptylineはセロトニン取り込みとノルアドレナリン取り込みに対するIC_50値が近似していると報告されているが,今回の実験においてGAT1,GAT3とSETに対するIC_50値が近似していた。これらの結巣から,amitriptylineの臨床的抗うつ効果はモノアミンと同時にGABA取り込み阻害作用が関与している可能性がある。
The clinical effectiveness of antidepressants was thoug}lt to be mainly due to their inhibitory effects on monoamine uptake, but the possible role of GABAergic neural transmission in affective disorders has been suggested by several experiments. We examined the effects of 11 antidepressants on the activities of three GABA transporter sub types (GAT1, GAT2 and GAT3) serotonin transporter or a glial glutamate transporter expressed in COS-7 cells. All the antidepressants studied dose-dependently inhibited the activities of serotonin transporter. Desipramine,nortripty line, amitriptyline and maprotiline inhibited certain type of GABA transporters,while the other antidepressants had negligible effects. Amitriptyline,which was reported to have a similar IC_50 value for serotonin uptake to noradrenaline uptake,showed similar IC_50 values for GAT1, GAT3 and SET in the present study. These results suggested that clinical antidepressive effects of amitriptyline might reflect the inhibition of GABA uptake as well as monoamine uptake.
