Studies on regeneration of residual pancreas after partial pancreatectomy

Present study was undertaken to reveal the effect of partial pancreatectomy and pre and postoperative administration of synthetic trypsin inhibitor on residual pancreatic regeneration in rats. Splenic and gastric of the pancreas in young wistar strain rats weighing about 200g were resected yealding about 60% pancreatectomy. Synthetic trypsin inhibitor, N.N. Dimethylcarbamoylmethyl 4- ( 4- quanidinobenzoyloxy) phenylacetate methane sulfonate, FOY 305, was dissolved into drinking water and given ad lib from 7th preoperative day. After partial pancreatectomy the ornithine decarboxylase activity, putresine content and mitotic index of residual pancreas increased and were accompanied by increased pancreatic weight to reveal regeneration. The increasing rate was enhanced by administratim of trypsin inhibitor. Chronological changes in mitotic indices and cell size of acinar cells and islet cells suggested that regeneration mode of both acinus and islet after pancreatectomy was hyperplasia at the early stage and hypertrophy at the late stage and that hyperplasia was more potent to pancreatic regeneration that hypertrophy. Trypsin inhibitor enhanced both hyperplasia and hypertrophy of acinar cells but affected only hypertrophy of islet cells slightly. Increased amylase content of residual pancreas showed that regenerated pancreas had normal exocrine function.
