Jerneのネットワーク仮説より,あるイディオタイプに対応する抗イディオタイプ抗体の超可変部は抗原と相同の構造を有するという事実が導きだされ,従来の抗原では免疫応答の惹起できなかった腫瘍分野でこれを抗原代替物とするワクチン療法が試みられている。私共は今回,種属間共通腫瘍関連抗原を認識する単クローン抗体に対して作成した単クローン抗イディオタイプ抗体を用い,Fisher ラット由来9Lグリオーマ系に対するワクチン療法実験を行なった。その結果,抗イディオタイプ抗体の免疫により,元の抗原と反応する抗抗イディオタイプ抗体が産生され,更に遅延型過敏反応の惹起,in vitro sensitizationによるヘルパーT細胞及び細胞障害性T細胞の誘導という細胞性免疫の積極的関与を示唆する結果が得られた。そして,抗イディオタイプ抗体を予め免疫することにより,in vivoで有意な腫瘍増殖抑制効果が生じることも確認された。以上の結果より,自己抗原に対する免疫不応という障害に阻まれていた腫瘍免疫療法に対し,抗イディオタイプ抗体の応用が一つの突破口となりうる可能性が示唆された。
According to the Jerne's network theory, an internal idiotope image exists for each antigen, and a paratope of antiidiotypic antibody (Ab2) which is complementary to the idiotope of an antibody (Ab1) can mimic the structure of the antigen. Therefore, an Ab2 might elicit immune responses when inoculated as a surrogative antigen ('vaccine'), by up regulating specific tolerized idiotypic cascade. Several works with such surrogative antigens as tumor vaccines have been achieved with variable success. Herein we have analyzed the induction of tumor-specific immunity related to a idiotope associated with a monoclonal anti- 'tumor associated antigen' antibody in a syngeneic 9L gliosarcoma inplantation model. An antiidiotopic monoclonal antibody (Ab2) was made against the paratope of Ab1, and immunization of Fisher 344 rats with Ab2 induced specific immune responses listed below. 1) generated anti-Ab2 sera (polyclonal Ab3) : Ab3 could bind to the original antigen, which suggested that Ab3 share the structure of paratope of Abl, 2) upregulated T cell-mediated immune responses: Delayed type hypersensitivity (DTH) in Fisher 344 rat was induced when primed with KLH-Ab2 and challenged with 9L cells. In vitro sensitization with inactivated 9L cells could generate significantly high blastgenesis of Ab2-immunized rat spleen cells (SPC's) than control groups of SPC's. It also induces specific killer activity of Ab2-immunized SPC's against 9L cells. 3) inhibited the tumor growth in vivo : The growth of inplanted tumorwas significantly inhibited in Ab2 pre-immunized group. These results suggest the possible regulation of tumor-specific idiotypic humoral and cellular cascades induced by immunization with Ab2 as a tumor vaccine.
