To clarify the role of autonomic nervous system in digitalis-induced arrhythmia, effect of ouabain on sympathetic activity was studied by measuring amplitude and frequency of impulses taken from the sympathetic, aortic and vagal nerves in rabbits. 40mcg/kg of ouabain was administered intravenously every 10 minutes, and 19mcg/kg of ouabain was given intracisternally (i. c.) or intralateraloventricularly (i. l. v.). Plasma catecholamine was measured simulataneously with the measurement of sympathetic activity. In the control rabbits, intravenous administration of ouabain caused arrhythmias without apparent sympathetic hyperactivity. In the denervated rabbits in which the 9th and 10th cranial nerves were transsected, sympathetic activity was increased by 75% and development of arrhythmia was slightly facilitated. The i. c. and i. l. v. administrations of ouabain developed prominent increases in sympathetic activity by 238% (i. c.) without arrhythmia and 447% (i. l. v.) with transient arrhythmias, respectively. Sympathetic activity was closely related to plasma catecholamine concentration in each experimental condition. Activity of the aortic nerve was slightly increased without apparent hemodynamic changes and its afferent impulse suppressed sympathetic activity. These results lead to the following conclusions : 1. Sympathetic activity might be unessential for the appearence of ouabain-induced arrhythmia, which was provoked by a direct action of digitalis on the heart, sympathetic activity was suggested to play only an accelerative role for the occurrence of the arrhythmia. 2. Sympathetic activity was closely related to plasma catecholamine concentration. 3. Enhanced activity of the aortic nerve which might be chemically sensitized by ouabain suppressed the sympathetic nervous system.
